For bacteria, snipping apart DNA that bears certain signature sequences is a defense mechanism. For scientists working in the lab, the same strategy can be a powerful research tool. With a newly discovered component of an adaptive bacterial immune system, scientists have identified a targeted method of slicing DNA that they say can be easily customized for a variety of applications in the lab.

Tools that snip apart DNA strands in defined locations are essential for editing genomes in the laboratory to study or alter gene function. To target the specific site in the genome they are interested in, researchers often have to design and produce a protein that will recognize and bind to that particular DNA sequence, a laborious and time-consuming process.

But this could change, says Howard Hughes Medical Institute investigator Jennifer Doudna at the University of California, Berkeley. In a paper published online June 28, 2012, in the journal Science, she and her collaborator Emmanuelle Charpentier of the Laboratory for Molecular Infection Medicine at Ume University in Sweden report the discovery of an RNA-based complex used by bacteria to guide the DNA-cutting enzyme Cas9 to specific sites in the genomes of viruses and other invaders, thus silencing their genes. From this bacterial complex, Doudna and her colleagues have crafted a system with which an easily programmable guide RNA can be used in the lab to direct Cas9 to cleave double-stranded DNA at a desired target sequence.

"We can direct it to any site we select," says Doudna. "Because the guide RNA contains both the structure required for Cas9 binding and a separate guide sequence that can base pair with DNA, we can program Cas9 to cleave a specific site by simply changing the guide sequence. This system offers a straightforward way to cleave any desired site in a genome, which could be used to introduce new genetic information by coupling it to well-known cellular DNA recombination mechanisms."

Doudna, a biochemist, is interested in the molecular mechanisms by which RNA can influence gene expression. She says the discovery of an RNA-programmable DNA cleaving enzyme stems from a collaboration with Charpentier established last year. Both labs were studying different aspects of RNA-based defensive systems in bacteria that recognize and destroy the genomes of invading viruses and plasmids.

First described in the late 1980s, the system is called CRISPR, for Clustered Regularly Interspaced Short Palindromic Repeats. In response to a viral infection or plasmid transformation, bits of the invader's DNA known as proto-spacers are integrated into the host chromosome. The captured sequences are transcribed and processed to form short crRNAs, which serve as RNA recognition elements that bind to corresponding sequences in foreign DNA. Guided by the RNAs, proteins known as Cas (CRISPR-associated) then move in and attack the invaders, cleaving their DNA and silencing them.

Researchers studying CRISPR systems in various bacteria had found that in most cases a single crRNA joins with a large, multi-protein complex to attack viruses and plasmids. However, Charpentier had discovered that in Streptococcus pyogenes, a human pathogen, crRNAs could only be produced in the presence of a second RNA, which they called a trans-activating crRNA (tracrRNA). In addition, the S. pyogenes and related CRISPR systems require just a single protein, Cas9, for immunity to viruses targeted by crRNAs.

Doudna's lab worked with the Charpentier lab to investigate how Cas9 and crRNAs function in this bacterial immune system. Martin Jinek, an HHMI Research Specialist in Doudna's lab, succeeded in purifying the Cas9 protein. Krzysztof Chylinski, a graduate student in the Charpentier lab who is located at the Max F. Perutz Laboratories at the University of Vienna, used that sample to show that Cas9 needed both crRNA and tracrRNA to guide and execute its attack.

"We then decided to test whether we could link these two RNAs into a single, chimeric RNA molecule," Doudna says. Combining the elements of the crRNA and tracrRNA that were necessary for Cas binding and DNA target recognition into a single molecule would make the system easier to manipulate for laboratory use, she explains. It worked: the result was a DNA-cleaving enzyme that can be programmed with a single RNA molecule to cleave specific DNA sites.

The next steps, Doudna says, are to test the single-RNA construct along with Cas9 to find out whether the RNA-programmed enzyme works in the cells of eukaryotic organisms, such as worms, plants, and humans. If that is successful, she anticipates many practical applications of the tool. For biotechnology efforts ranging from engineering biofuel-producing microorganisms to enabling cell-based medical therapies, "having a simple and inexpensive tool for genome editing available will be very important," she says.

European leaders seek roadmap for future growth

BRUSSELS (AP) ? Economic growth was the mantra among European leaders as they began a crucial summit on Thursday, though expectations of a breakthrough on the explosive issue of pooling government debt appeared to have fallen by the wayside.

European Commissioner for Economic Affairs Olli Rehn said he expected leaders would agree on new growth measures, as well as on action to reduce borrowing rates for Spain and Italy, which are approaching unmanageable levels.

But Merkel has been reluctant to expose her country to new costs, and is concerned eurobonds would ease the pressure on countries like Greece and Spain to reform their economies.

"I think we should stop talking about eurobonds now because, with the German government's 'no,' with this definitive 'no' from Mrs. Merkel, eurobonds are now a non-issue," the president of the European Parliament, Martin Schulz, said Thursday.

"I personally continue to see it as a good solution, a sensible one, but there is no sense in conducting theoretical debates when the house is on fire," Schulz told Germany's ZDF television.

While pressure on Merkel has been building, she is not alone in her opposition to have EU countries accrue joint debt. Swedish Prime Minister Fredrik Reinfelt said Thursday that what was needed was not mutual debt but more financial reforms by Europe's trouble countries ? measures he said would make them more competitive.

Dutch Prime Minister Mark Rutte, too, spoke forcefully Thursday against mutual debt.

"We only solve Europe's problems by respecting past agreements and by making markets function better. The only way out of this crisis is to help southern European nations, provided they accept responsibility and strict criteria," Rutte said.

Investors are looking for signs of a clear strategy to deal with the debt crisis. They are not hopeful, though, and sold off stocks across Europe on Thursday as European leaders prepared for the summit.

"The good news is that this time around expectations are very low; the bad news is that the main players seem to be diametrically opposed when it comes to a strategy for ending the crisis," said Gary Jenkins, managing director of Swordfish Research, a London-based consulting firm.

The French plan to stimulate growth, and thereby increase government tax revenues, is relatively modest. Although worth ?130 billion ($162 billion), it is expected to consist mostly of European funds already earmarked for development.

Italy's Prime Minister Mario Monti, at risk of losing his job because of voter frustration with budget cutbacks, said Wednesday in Brussels that Italians have made great sacrifices and gotten their country's deficit under control but the yields on Italian debt have soared to one-year highs anyway.

If Italians become discouraged, Monti warned, of the possible rise of "political forces which say 'let European integration, let the euro, let this or that large country go to hell', which would be a disaster for the whole of the European Union."

Monti said he's ready to work until Sunday night ? instead of the scheduled Friday end of the summit ? to ensure that leaders produce a growth package convincing enough to calm financial markets.

Spain's prime minister is sounding especially desperate.

"The most urgent issue is financing," Mariano Rajoy said. "We can't continue for a long time to finance ourselves with these prices; there are many institutions and financial entities that don't have access to financial markets."

Simon Tilford of the Center for Economic Reform said, "We're seeing the French, Italians and Spanish showing a greater readiness to act as one."

In the past, they were reluctant to isolate Merkel, he said.

"But that flexible approach ... has delivered very little. They have grown alarmed and frustrated," he said. "If anyone is to lead the charge, it may be Monti, he is the one who has the most credibility on the European stage" ? and the most to lose if pressure on Merkel fails.

While France has been the traditional partner ? and counterweight ? to Germany in European dealings in the past, President Francois Hollande has just seven weeks of governing under his belt, and built his career as a consensus-builder. And his country's economy is weaker, with growth forecast at just 0.4 percent this year.

Hollande was grinning broadly Wednesday night at the one concession he has been able to wring from Merkel so far, an agreement to put growth on the European agenda alongside austerity measures.

Merkel, standing stiffly at Hollande's side ahead of bilateral talks in Paris, agreed to push for the ?130 billion ($162 billion) stimulus package that Hollande has vaunted, even though it is largely just a re-packaging of existing EU funds.

Even if leaders of all 26 other EU nations line up against Merkel, she cannot bend very far.

She needs the German Parliament to approve the eurozone's permanent rescue fund, the European Stability Mechanism, and a European budget-discipline pact, both expected to happen Friday. And many measures floated as possible solutions could require changes to Germany's constitution.

Sony made a big mistake when they released the PS3 with a ?500 RRP just a year after Microsoft had launched the Xbox 360 for ?280. Sony?s initial struggles were largely a result of their failure to convince consumers that their console was significantly better than Microsoft?s. As a result they lost out on a number of exclusives due to the 360?s much larger install base. While sales of the PS3 have since picked up pace, especially in Europe, Sony would have been in a much healthier position if their launch had been stronger. It is then, vitally important for both companies to get the price point right first time round or risk failing to entice consumers to invest in new hardware.

Release date

For Microsoft, releasing a whole year earlier than the PlayStation 3 proved to be a big advantage. When they released the Xbox 360 they had no competition, with the PlayStation 2 nearing the end of its life cycle, this meant that consumers only had one choice to make, whether to buy an Xbox 360 or buy nothing at all. For Sony this caused many issues when they released the PS3, many gamers had already invested in an Xbox 360 and were not willing to spend a significant amount of money on yet another console. On top of this, Microsoft had already built up a solid library of games and had many more ready for release, making their product far more attractive. Something that they will both have to also keep in mind when deciding upon a release date is when making sure that there are no major current generation games being released around the same time, as this would devalue the need for buying a new console altogether. If Rockstar release the next Grand Theft Auto on current systems, it could be argued that they still have plenty to offer.

Games

Having an attractive selection of games is a big selling point for a console, if you give gamers titles which have unique features which can only be run on a new console then you give them a reason to go out and buy it. Making existing games for current gen consoles compatible with next gen machines is also important. If consumers know that they can still play the games they already own on the new system, there will be less hesitation in moving on. Shortly after Microsoft launched the Xbox 360, they had big games like Halo 3 and new franchises like Gears of War available to consumers. On the other hand Sony had several great games, such as Resistance: Fall of Man and Motorstorm, but didn?t have any major titles to drive sales.

So it is clear that both Sony and Microsoft are going to have to make sure they consider a number of different factors when releasing new hardware. They will need to ensure that they have a unique selling point to make their products attractive to the consumer. Sony especially simply cannot afford to get it wrong a second time round as the company is already struggling to cope with slumping sales in its other product lines. While Microsoft will be desperate to continue the momentum gained by the Xbox 360.

Scientists who search for new pesticides for use in humanity's battle of the bugs and other threats to the food supply have been learning lessons from Mother Nature, according to a new analysis. It concludes that more than two out of every three new pesticide active ingredients approved in recent years had roots in natural substances produced in plants or animals. The article appears in ACS' Journal of Natural Products.

Charles L. Cantrell and colleagues point out that there have been many analyses of the impact of natural products -- substances produced by living plants, animals and other organisms -- on the production of pesticides. None, however, has ever looked at the impact of natural products and natural product-based pesticides in fostering new active ingredients (NAIs) in pesticides on the U.S. market, based on NAI registrations with the U.S. Environmental Protection Agency. The scientists filled that information gap with results that they say defy conventional wisdom that natural products may not be the best sources for NAIs.

The analysis found that between 1997 and 2010, more natural products were registered as NAIs for conventional pesticides and biopesticides than any other type of ingredient. The authors report that when biological ingredients and natural products recreated in labs are included, more than 69 percent of all NAIs registered in that time frame have natural origins.

The authors acknowledge funding from the U.S. Department of Defense through the Armed Forces Pest Management Board.

Night one in Las Vegas was clearly a ball of nerves for several acts. Did the judges' favorites on America's Got Talent overcome their nerves to perform at their best on Tuesday?

Magic - Mind Reader Eric continued with his successful act by finding Sharon's first crush. While he managed to get "Robin" right, Sharon made up the name. Taylor Reed made a helicopter appear, but it was the same as the motorcycle he auditioned with down to the flashlight. Justin Rivera had a moving box sequence that was so obvious that it wasn't funny.

The Father/Daughter street singers had the dad forget a few words and dad was off key. Father/Young Daughter was chipmunky, pitchy, and painful.

Comedians - With six comedians, youngest Jacob Williams had strong material that Howard didn't like. Sammy Obeid made some girlfriend knocks, Dave did impersonations, and Kellen did a psychic/robbery joke that was too slow. Tom Cotter continued the "that's their name" jokes and the "I want to kill my ex" jokes. Frank Roche took a bad gamble and choked; Howard mercy killed him.

Advanced: Jacob and Tom

Kids - As a reminder to the audience children still have to do homework. Yay child labor laws. Sebastien was still strong with his mariachi band and didn't need a mic. Issac Brown was pitchy and pretty bad; I get that he's a fan of the Jackson 5, but he really needs suitable material. The Untouchables (aka Miami All-Stars Jr.) had a disco act that wasn't bad; it showed range. Amazing Elisabeth continued to be amazing and out of breath; the act would be better with a woman, but this girl showed promise. Edon did a really good job, considering he was the only kid playing an instrument.

Advanced: Edon, Sebastien, and The Untouchables

Male Singers - Ulysses, the often-sleeping theme song singer, sang "Secret Agent Man" but his pronunciation of "agent" was odd. Howard still didn't get it. Daniel Park, Jake Rogers, and questionable Sargent Tim Poe had small forgettable moments. Tim Hockenberry had that wonderful Joe Cocker gruff as he sung "Maybe I'm Amazed."

Advanced From Day One: The thing that didn't surprise me was that most if not all of the novelty acts made it through (light/sand/drag/nuts/dog/iLuminate knock-off) as did both bike/stunt danger teams. I am happy to announce that the overdressed Lion King act and the underdressed Loyalty were both eliminated. If that means we have to hold on to 787 and All That!, then I'm okay.

As a future note, find the right costuming for whatever category you're in. Only the Aussie girl managed to survive in the female category. In an utterly happy move, none of the classical males survived. No more of Andrew's tears through his ice blue contacts.

It seems like about 25 of the spots have been taken, so the remaining acts will have to fight for what's left tomorrow.

2012 Atlanta Falcons Offense:
The Falcons thought they were one player away from Super Bowl contention when they traded a barrage of picks for Julio Jones in the 2011 NFL Draft. They clearly misevaluated their talent, as they discovered when the Giants essentially blanked them on the scoreboard in January.

That's not to say that Jones didn't help their offense. Matt Ryan threw for career-highs in both passing yards (4,177) and touchdowns (29) even though he attempted five fewer passes than he did in 2010. This includes a game-high seven receptions for 64 yards in the aforementioned defeat against New York.

Jones will emerge as Ryan's No. 1 target in 2012. His sheer talent isn't the only reason; the decline of Roddy White is also a factor. White is still a receptions machine - he has 215 catches the past two years - but he's lost some of his explosiveness, which is hardly a surprise because he'll turn 31 in November. White also has unreliable hands; no one in the NFL had more drops than White last season (15).

Elsewhere in Ryan's receiving corps, Harry Douglas and Tony Gonzalez have returned despite being a free agent and a retirement candidate, respectively. Douglas is an underrated slot receiver, while Gonzalez, 36, rebounded last year from a dismal 2010 campaign in which he looked unbelievably slow. Unfortunately, the future Hall-of-Famer is playing on his last legs, and his talent could completely erode by September. Atlanta did not draft his successor, so it'll have to do so next April.

One of the reasons why the Falcons were criticized for giving up too much for Jones was because they depleted their depth and prevented themselves from addressing a very important position of need. That would be left tackle, where Sam Baker and Will Svitek took turns embarrassing themselves last season. Atlanta could have chosen Riley Reiff with the No. 22 pick in the 2012 NFL Draft, but instead had to settle for Lamar Holmes in the third round. Holmes has some athleticism at 6-5, 325, but he's raw and won't be able to contribute immediately.

The rest of the offensive line is looking pretty solid. The Falcons spent their second-round selection on Wisconsin's Peter Konz, a borderline first-round prospect. Konz will start at right guard this season but will eventually move to center to succeed Todd McClure, who played well at 34 last year. Left guard Justin Blalock and right tackle Tyson Clabo are both excellent in pass protection, though the latter needs to cut down on his penalties (eight in 2011).

Another position the Falcons couldn't address because of a lack of resources was running back. Michael Turner had a solid 2011 campaign overall, but really looked like he was running on fumes down the stretch. He averaged more than 3.6 yards per carry in only one of his final seven games, and that came against a Buccaneer team that quit. Head coach Mike Smith told the media that Turner will be featured less this year; some of his touches will go to Jacquizz Rodgers, a very small (5-6, 196) but speedy running back whom the Falcons acquired in the fifth round of the 2011 NFL Draft. Rodgers cannot carry a full workload, however, so Atlanta will have to find a complementary player next April.

2012 Atlanta Falcons Defense:
The Falcons have obtained exactly one big-name free agent in each of the previous two offseasons. They'll be hoping that this year's acquisition, Asante Samuel, is better than 2011's Ray Edwards.

Samuel was a steal for a late-round pick. He was disgruntled and highly paid in Philadelphia, so Andy Reid was more than happy to part ways with him. Samuel turned 31 in January, but still played extremely well last year, surrendering a sterling completion percentage of 47.5. That's almost as good as Brent Grimes' unbelievable figure (44.3), so the two should form an incredible cornerback tandem. Grimes was missed at the end of last season when he went down with a knee injury. Meanwhile, Dunta Robinson, who really struggled in 2011, will move to a more comfortable slot corner position.

The one weakness in Atlanta's secondary in the wake of the Samuel trade is free safety Thomas DeCoud, who was often picked on by opposing quarterbacks last season. Strong safety William Moore made up for it, but he needs to stay healthy. He's missed 18 games in his 3-year career, which does not include the aforementioned loss to the Giants; he barely played in that contest because of a groin injury he suffered in the first quarter.

As for Edwards, the former Viking really disappointed. He could barely generate any pressure on the quarterback, notching just 3.5 sacks despite playing across from John Abraham and his 9.5 sacks. Both players were very good in run support, but Edwards really has to pick up the slack in 2012, especially considering that Abraham, at 34, is past his prime and could be in for a major decline any season. The Falcons spent a fifth-round selection on Jonathan Massaquoi, but he probably won't contribute very much this year.

The interior of the defensive front is pretty solid. Jonathan Babineaux is a very well-rounded player; he can apply some pressure on the quarterback and hold his own against the run. Corey Peters, meanwhile, did an OK job in run support last year, but hardly ever rattled opposing signal-callers. Atlanta will want 2009 first-round pick Peria Jerry to push Peters, but it appears as though that ship has sailed. Jerry hasn't been the same since shredding his knee during his rookie campaign.

The Falcons had one major loss this offseason, and that was Curtis Lofton, the team's leading tackler (147 tackles), who signed with arch rival New Orleans. They added Lofa Tatupu as a possible replacement. He'll battle it out with 2011 third-rounder Akeem Dent to see who lands the starting gig. Tatupu was out of the league last year, so it's tough to speculate what he could possibly contribute. Dent, meanwhile, barely played as a rookie, so he's just as much of an unknown.

Surrounding the winner of the Tatupu-Dent competition are Sean Weatherspoon and Stephen Nicholas. Weatherspoon didn't play very well as a rookie in 2010, but really stepped up last year, becoming one of the top linebackers in the NFL. Nicholas is a pretty decent two-down run-defender.

2012 Atlanta Falcons Schedule and Intangibles:
In four seasons, Matt Ryan is 26-5 in the Georgia Dome. That's an impressive record on paper, but it didn't seem to matter much when the Packers waltzed into the Falcons' house and slaughtered them, 48-21, last January.

Eric Weems is a very good punt returner. He didn't score last year, but maintained a 9.8 average, while the Falcons restricted the opposition to 4.8. Weems was outgained on kickoffs, however. No touchdowns there either, for or against.

Matt Bryant has been one of the top kickers in the NFL over the past two years. He was 28-of-31 in 2010 and 27-of-29 last season. He's 3-of-3 from 50-plus in that span. He just turned 37.

Matt Bosher didn't have a successful rookie campaign as Atlanta's new punter. He averaged 42.7 yards per attempt with 27-of-70 tries inside the 20.

Atlanta's second-place schedule is pretty difficult. The team has to deal with the Chiefs (road), Broncos (home), Chargers (road), Eagles (road), Cowboys (home), Giants (home), Lions (road), as well as the Saints and Panthers twice.

Goals Entering the 2012 NFL Draft: The offensive line is the greatest area of concern. Left tackle and right guard are major problem areas. A successor for Todd McClure is also needed at center. Unfortunately, Atlanta doesn't have a first-round pick.

2012 NFL Draft Accomplishments: This is a tough grade because the Falcons didn't pick until No. 55 and had only two selections in the top 150. I loved what they did with their first choice in picking Peter Konz. The Wisconsin lineman fell because of injury concerns, which were overblown. He'll be a force on Atlanta's front, starting out at guard and then taking over for McClure in a year or two.

The rest of Atlanta's picks were decent, save for Lamar Holmes, who was not in the top 225 of many major big board. He was a big reach at No. 91, but maybe the Falcons, who didn't pick until No. 157, thought there would be a good chance he wouldn't be available in the fifth round.

Ovie Mughelli is coming off a torn MCL, so it's understandable why the Falcons would draft a fullback. I think this is a bit early for Bradie Ewing, but it's not much of a reach.

This is a really good value pick. Jonathan Massaquoi was ranked No. 84 on Mike Mayock's big board, and he was No. 116 on the consensus rankings. The Falcons had to find a young pass-rusher to perhaps eventually take over for John Abraham.

192. Charles Mitchell, SS, Mississippi State B Grade
Safety depth had to be addressed, so this pick makes sense. Charles Mitchell could have gone a bit later than this, but given that this is Atlanta's penultimate pick, it's not a big deal.

249. Travian Robertson, DT, South Carolina B Grade
The Falcons don't have much depth at defensive tackle, so this pick makes sense. Travian Robertson probably should have been selected at this spot.
